Background Acute lung injury (ALI) is accompanied by DNA damage and repair of epithelial cells, but the DDR of macrophages and its? role in regulation of inflammatory response remains largely unknown. we aim to elucidate the regulatory role of DDR signaling in macrophages in the airway inflammation of ALI and its? possible mechanism, using primary cultured macrophages and LPS-induced ALI model, thus providing theoretical basis for pharmacologic therapy of ALI.
Materials and methods We used ku-55933 to block ATM both in the in-vitro macrophages cell line and the in-vivo ALI mice model.
Results We observed that treatment with different concentrations of ATM inhibitors ku-55933 reduce LPS-induced expression and secretion of inflammatory cytokines in RAW264.7 cells. Similarly, the pretreatment group with chemical inhibition of DDR produced an alleviated inflammatory response compared with the ALI group.
Conclusion Inhibition of macrophage ATM function has a protective effect on LPS-induced ALI inflammation, and ATM is involved in LPS-induced DNA damage and repair.
